From the episode

Think of Kyle Williams' best game as a pro against the Bucs last year. And remember what routes he caught? Over routes when he went in motion. But the guy obviously can't win vertically like that.

Williams misaligned; should be Z receiver, not slot over Douglas.

Kyle Williams is misaligned in the Eagles' offense. His best production comes on over routes when in motion (like his best game against the Bucs last year), but he can't win vertically. Matt Harmon argues Williams should be the Z receiver, not the slot, and should be used over Pop Douglas given his skill set and the scheme's limitations.
